Separa\u010dn\u00fdm modulom v on-line zapojen\u00ed je nano syst\u00e9m HPLC s chromatografiou na \u010dipe. Tak\u00e9to spojenie pr\u00edstrojovej techniky umo\u017e\u0148uje ve\u013emi r\u00fdchlu, citliv\u00fa a presn\u00fa identifik\u00e1ciu jednotliv\u00fdch peptidov pri pou\u017eit\u00ed minim\u00e1lneho mno\u017estva vzorky. Dynamick\u00fdm rozsahom 5-tich poriadkov umo\u017e\u0148uje stanovi\u0165 ved\u013ea seba stopov\u00e9 analyty v pr\u00edtomnosti vysoko zast\u00fapen\u00fdch zl\u00fa\u010den\u00edn. Svojim vysok\u00fdm rozl\u00ed\u0161en\u00edm, r\u00e1dovo 40 000, zamedzuje falo\u0161ne pozit\u00edvnym identifik\u00e1ci\u00e1m. V\u010faka technol\u00f3gii Jet Stream m\u00e1 vy\u0161\u0161iu citlivos\u0165 merania oproti klasick\u00fdm QQQ a spolu s vysoko citliv\u00fdm detektorom umo\u017e\u0148uje merania aj v ultrastopovej oblasti. Tento typ hmotnostn\u00e9ho spektrometra sa naj\u010dastej\u0161ie vyu\u017e\u00edva pre kvantitat\u00edvne anal\u00fdzy. Separa\u010dn\u00fdm modulom v on-line zapojen\u00ed je analytick\u00fd syst\u00e9m HPLC. Tak\u00e9to spojenie pr\u00edstrojovej techniky umo\u017e\u0148uje kvantifik\u00e1ciu vopred vyselektovan\u00fdch peptidov vo vzorke. M\u00e4kk\u00e1 ioniz\u00e1cia sa vyu\u017e\u00edva pre \u0161t\u00fadium \u0161irok\u00e9ho spektra l\u00e1tok zah\u0155\u0148aj\u00facich biomolekuly (prote\u00edny, peptidy, sacharidy, nukleov\u00e9 kyseliny atd.) Na \u00fastave sa vyu\u017e\u00edva na identifik\u00e1ciu prote\u00ednov prostredn\u00edctvom ur\u010denia ich hmotnosti, hmotnosti ich jednotliv\u00fdch peptidov a ich fragmentov. Nameran\u00e9 hmotnostn\u00e9 spektr\u00e1 sa n\u00e1sledne porovn\u00e1vaj\u00fa so svetov\u00fdmi datab\u00e1zami hmotnostn\u00fdch spektier prote\u00ednov. V\u00fdsledkom je identifik\u00e1cia analyzovan\u00e9ho prote\u00ednu. Na automatiz\u00e1ciu nan\u00e1\u0161ania vzorky na MALDI ter\u010d\u00edk sl\u00fa\u017ei zbera\u010d frakci\u00ed Bruker Proteineer fcII, ktor\u00fd je spojen\u00fd so separa\u010dnou technikou vysoko \u00fa\u010dinnej kvapalinovej chromatografie \u2013 HPLC (obr. 4). Po HPLC separ\u00e1cii sa oddelen\u00e9 peptidy frak\u010dn\u00fdm kolektorom nan\u00e1\u0161aj\u00fa na MALDI ter\u010d\u00edk. Na pr\u00edpravu vzorky tkan\u00edv, ktor\u00e9 maj\u00fa by\u0165 podroben\u00e9 MALDI anal\u00fdze, sa pou\u017e\u00edva pr\u00edstroj Bruker ImagePrep. Pr\u00edstroj rovnomerne nan\u00e1\u0161a extr\u00e9mne tenk\u00e9 vrstvy kry\u0161t\u00e1likov matrice a pomocou optick\u00fdch senzorov kontroluje stav nanesenej matrice, jej homogenitu a hr\u00fabku vrstvy. Tak\u00e1to kontrola vrstvy nanesenej matrice je nevyhnutn\u00e1 pre dosiahnutie \u010do najlep\u0161ieho rozl\u00ed\u0161enia pri MALDI meran\u00ed.
