N\u00e1dorov\u00e9 ochorenia predstavuj\u00fa obrovsk\u00fd zdravotn\u00fd, psychick\u00fd a soci\u00e1lny probl\u00e9m pre pacientov, ich okolie
i cel\u00fa spolo\u010dnos\u0165. Zmena \u017eivotn\u00e9ho \u0161t\u00fdlu, prostredia a n\u00e1rast vekov\u00e9ho priemeru \u013eudskej popul\u00e1cie sp\u00f4sobuj\u00fa, \u017ee po\u010det ochoren\u00ed a nov\u00fdch onkologick\u00fdch pr\u00edpadov sa neust\u00e1le zvy\u0161uje. V\u00fdskum realizovan\u00fd v CIB v oblasti nanomedic\u00edny rie\u0161i z\u00e1va\u017en\u00fd probl\u00e9m spojen\u00fd s chemoterapiou, konkr\u00e9tne s jej ne\u017eiad\u00facimi ved\u013eaj\u0161\u00edmi \u00fa\u010dinkami, a je zameran\u00fd na dva z\u00e1kladn\u00e9 pr\u00edstupy rie\u0161iace tento probl\u00e9m:<\/p>\n\n\n\n

  1. V\u00fdvoj selekt\u00edvneho nanotransportn\u00e9ho syst\u00e9mu pre cielen\u00fd transport protin\u00e1dorov\u00fdch lie\u010div.
    Hlavn\u00fdm cie\u013eom zamerania je v\u00fdvoj novej gener\u00e1cie transportn\u00fdch syst\u00e9mov schopn\u00fdch efekt\u00edvne pren\u00e1\u0161a\u0165 dostato\u010dn\u00e9 mno\u017estvo lie\u010div a vyzna\u010duj\u00facich sa silnou afinitou pre v\u00e4zbu so \u0161pecifick\u00fdmi receptormi nach\u00e1dzaj\u00facimi sa na povrchoch n\u00e1dorov\u00fdch buniek. Takto dizajnovan\u00fd transportn\u00fd syst\u00e9m lie\u010div m\u00e1 potenci\u00e1l by\u0165 efekt\u00edvne modulovate\u013en\u00fd aj pre mutovan\u00e9 n\u00e1dorov\u00e9 receptory jednotliv\u00fdch pacientov, \u010d\u00edm m\u00f4\u017ee prispie\u0165 k personalizovanej lie\u010dbe n\u00e1dorov\u00fdchochoren\u00ed.<\/li>\n\n\n\n
  2. Fotodynamick\u00e1 terapia rakoviny (PDT)
    Hlavn\u00fdm cie\u013eom tohto zamerania je popis mechanizmov bunkovej smrti vyvolanej fotodynamickou akciou a na jeho z\u00e1klade n\u00e1vrh zefekt\u00edvnenia PDT na bunkovej \u00farovni. N\u00e1sledne, na z\u00e1klade poznatkov z\u00edskan\u00fdch na bunkovej \u00farovni, navrhn\u00fa\u0165 protokoly PDT na \u00farovni modelov\u00e9ho syst\u00e9mu CAM (membr\u00e1na kurac\u00edch embry\u00ed), \u00farovni in vivo (zvieracie modely) s mo\u017enou aplik\u00e1ciou v selekt\u00edvnej lie\u010dbe n\u00e1dorov\u00fdch ochoren\u00ed u \u013eud\u00ed.<\/li>\n<\/ol>\n\n\n\n

    Prote\u00ednov\u00e9 in\u017einierstvo<\/strong>
    V\u00fdvoj a v\u00fdskum prote\u00ednov pou\u017e\u00edvan\u00fdch pri biologickej lie\u010dbe<\/strong>
    V\u00fdskum v tejto t\u00e9me je orientovan\u00fd na v\u00fdvoj a charakteriz\u00e1ciu prote\u00ednov a enz\u00fdmov s nov\u00fdmi vlastnos\u0165ami, vhodn\u00fdmi pre medic\u00ednske, diagnostick\u00e9 a biotechnologick\u00e9 aplik\u00e1cie. V\u00fdvoj nov\u00fdch prote\u00ednov\/enz\u00fdmov s po\u017eadovan\u00fdmi vlastnos\u0165ami bude zalo\u017een\u00fd na vyu\u017e\u00edvan\u00ed \u201estate-of-the-art\u201c techn\u00edk racion\u00e1lneho dizajnu mut\u00e1ci\u00ed v t\u00fdchto biomakromolekul\u00e1ch a tzv. evolu\u010dn\u00fdch met\u00f3d, ako s\u00fa displej (ribozom\u00e1lny a kvasinkov\u00fd) technol\u00f3gie. \u0160peci\u00e1lny d\u00f4raz bude
    kladen\u00fd na v\u00fdskum a v\u00fdvoj prote\u00ednov na b\u00e1ze DARPinov a monoklon\u00e1lnych protil\u00e1tok, ktor\u00e9 s\u00fa \u00faspe\u0161ne pou\u017e\u00edvan\u00e9 pri biologickej lie\u010dbe pacientov. Tieto prote\u00edny s\u00fa schopn\u00e9 \u0161pecificky a s vysokou afinitou rozpozn\u00e1va\u0165 charakteristick\u00e9 epitopy komplexn\u00fdch prote\u00ednov, napr. \u0161pecifick\u00e9 receptory na rakovinov\u00fdch bunk\u00e1ch, ako aj jednoduch\u00fdch an\/organick\u00fdch ligandov. Novovytvoren\u00e9 prote\u00edny bud\u00fa n\u00e1sledne vyu\u017e\u00edvan\u00e9 na diagnostick\u00e9 a medic\u00ednske \u00fa\u010dely, napr. detekcia \u0161pecifick\u00fdch receptorov na tkaniv\u00e1ch a cielen\u00fd transport lie\u010div do postihnut\u00e9ho tkaniva.<\/p>\n\n\n\n

    <\/a>Starnutie a oxida\u010dn\u00fd stres<\/strong>
    V\u00fdskumn\u00e9 zameranie \u201eStarnutie a oxida\u010dn\u00fd stres\u201c v sebe zah\u0155\u0148a dve vz\u00e1jomne sa dop\u013a\u0148aj\u00face oblasti: molekulov\u00fa bioenergetiku bunkov\u00e9ho d\u00fdchania a bioenergetiku bunky a oxida\u010dn\u00fd stres. V\u00fdskum v t\u00fdchto oblastiach prin\u00e1\u0161a v s\u00fa\u010dasnosti mnoh\u00e9 poznatky, ktor\u00e9 m\u00f4\u017eu prispie\u0165 k objasneniu fundament\u00e1lnych ot\u00e1zok t\u00fdkaj\u00facich sa procesu starnutia a sn\u00edm s\u00favisiacimi mechanizmami vzniku a priebehu mnoh\u00fdch z\u00e1va\u017en\u00fdch ochoren\u00ed (predov\u0161etk\u00fdm n\u00e1dorov\u00e9 ochorenia a neurodegenerat\u00edvne
    ochorenia).<\/p>\n\n\n\n

    <\/a>\u017divotn\u00e9 prostredie<\/strong>
    V tejto t\u00e9mesa zaober\u00e1me v\u00fdvojom spo\u013eahliv\u00fdch a citliv\u00fdch met\u00f3d na detekciu ve\u013emi n\u00edzkych koncentr\u00e1ci\u00ed lie\u010div v biologickom prostred\u00ed, ako aj polutantov \u017eivotn\u00e9ho prostredia vo vode a potravin\u00e1ch. Zameriavame sa na v\u00fdvoj nano-senzorov zalo\u017een\u00fdch na modifikovan\u00fdch kovov\u00fdch povrchoch (predov\u0161etk\u00fdm strieborn\u00fdch a zlat\u00fdch) vhodn\u00fdch na detekciu n\u00edzkych koncentr\u00e1ci\u00ed molek\u00fal met\u00f3dou povrchovo zosilnenej Ramanovej spektroskopie (SERS). V\u00fdskum v tejto oblasti je \u00fazko koordinovan\u00fd s \u010dinnos\u0165ou startupu SAFTRA Photonics s.r.o, ktor\u00fd z\u00edskal v roku 2017 v\u00fdznamn\u00fd medzin\u00e1rodn\u00fd projekt v r\u00e1mci v\u00fdzvy SME – H2020. Tento projekt je venovan\u00fd v\u00fdvoju a optimaliz\u00e1cii nano-\u010dipu na detekciu n\u00edzkych koncentr\u00e1ci\u00ed organochloridov\u00fdch pestic\u00eddov vo vode a potravin\u00e1ch.<\/p>\n","protected":false},"excerpt":{"rendered":"
