\u010clenovia Katedry bunkovej biol\u00f3gie<\/strong><\/span> sa zameriavaj\u00fa na nasleduj\u00face oblasti v\u00fdskumu:<\/p>\n

Hyperic\u00edn, skyr\u00edn, fotodynamick\u00e1 terapia a mnohopo\u010detn\u00e1 liekov\u00e1 rezistencia n\u00e1dorov:<\/strong><\/span>
V\u00fdskum je zameran\u00fd predov\u0161etk\u00fdm na experiment\u00e1lnu onkol\u00f3giu. Na modeli n\u00e1dorov\u00fdch bunkov\u00fdch l\u00edni\u00ed, sf\u00e9roidov vytvoren\u00fdch z n\u00e1dorov\u00fdch bunkov\u00fdch l\u00edni\u00ed a mikron\u00e1dorov kreovan\u00fdch na chorioalantoickej membr\u00e1ne embry\u00ed prepelice japonskej sk\u00famame vplyv hyperic\u00ednu a skyr\u00ednu na angiogen\u00e9zu, prote\u00edny zapojen\u00e9 do priestorovej organiz\u00e1cie a adhez\u00edvnosti, ako aj schopnos\u0165 modulova\u0165 ve\u013ekos\u0165 a vlastnosti rezistentnej, tzv. bo\u010dnej popul\u00e1cie buniek. V r\u00e1mci v\u00fdskumu sa zaober\u00e1me aj \u0161t\u00fadiom vplyvu polymorfizmov transportn\u00e9ho prote\u00ednu BCRP a hypoxie na akumul\u00e1ciu hyperic\u00ednu so zameran\u00edm hlavne na mechanizmus regul\u00e1cie ABC transportn\u00fdch prote\u00ednov.<\/p>\n

DNA \/RNA aptam\u00e9ry:<\/strong><\/span>
V\u00fdskum sa zaober\u00e1 mo\u017enos\u0165ami uplatnenia r\u00f4znych funk\u010dn\u00fdch RNA molek\u00fal (aptam\u00e9ry, siRNA, a podobne) v personalizovanej medic\u00edne. Na\u0161ou snahou je zostavenie DNA-RNA nano\u010dast\u00edc, ktor\u00e9 v svojej \u0161trukt\u00fare maj\u00fa zabudovan\u00e9 viacer\u00e9 funk\u010dn\u00e9 RNA, tak\u00fdm sp\u00f4sobom, \u017ee k ich aktiv\u00e1cii d\u00f4jde len v n\u00e1dorov\u00fdch bunk\u00e1ch. Druhou l\u00edniou v\u00fdskumu je v\u00fdvoj RNA aptam\u00e9ru \u0161pecifick\u00e9ho vo\u010di SARS-CoV2 Spike prote\u00ednu.<\/p>\n

V\u00fdvojov\u00e9 procesy v mieche:<\/strong><\/span>
Na modelovom organizme laborat\u00f3rneho potkana sk\u00famame v\u00fdvinov\u00e9 procesy, ktor\u00e9 stoja za produkciou, migr\u00e1ciou a vz\u00e1jomn\u00fdmi interakciami jednotliv\u00fdch typov nervov\u00fdch a gliov\u00fdch buniek vo vyv\u00edjaj\u00facej sa mieche a pri\u013eahl\u00fdch \u0161trukt\u00farach v druhej polovici intrauterinn\u00e9ho v\u00fdvinu, po\u010das prv\u00fdch postnat\u00e1lnych t\u00fd\u017ed\u0148ov \u017eivota a v dospelosti u intaktn\u00fdch jedincov, ako aj v reakcii na minim\u00e1lne poranenie miechy. V\u00fdvin nervov\u00e9ho syst\u00e9mu \u0161tudujeme pomocou\u00a0 histologick\u00fdch a imunofluorescen\u010dn\u00fdch techn\u00edk, konfok\u00e1lnej a elektr\u00f3novej\u00a0 mikroskopie a molekul\u00e1rno-biologick\u00fdch anal\u00fdz. Samostatn\u00fa l\u00edniu v\u00fdskumu predstavuj\u00fa farmakokinetick\u00e9 anal\u00fdzy vybran\u00fdch substanci\u00ed in vivo<\/em>.<\/p>\n

S\u00fa\u010das\u0165ou laborat\u00f3ria je kompletn\u00e9 vybavenie potrebn\u00e9 na realiz\u00e1ciu experimentov in vitro<\/em> a ex ovo<\/em>: lamin\u00e1rne boxy (TELSTAR BIOSTAR, AURA 2000 M. Medzi \u010fal\u0161ie vybavenie laborat\u00f3ria spojen\u00e9 s kultiv\u00e1ciou a anal\u00fdzou buniek patr\u00ed automatick\u00fd analyz\u00e1tor po\u010dtu a viability buniek (Vi-Cell XR), automatick\u00fd mikroskop schopn\u00fd pracova\u0165 vo f\u00e1zovom kontraste a v du\u00e1lnej fluorescencii bez potrebnej interakcie \u010dloveka (IncuCyte) a kultiva\u010dn\u00e1 komora s regulovanou atmosf\u00e9rou (regul\u00e1cia CO2<\/sub>, O2<\/sub>, teploty a vlhkosti) (Coy Lab Products).<\/p>\n

Laborat\u00f3rium molekulovej biol\u00f3gie<\/strong><\/h4>\n

Laborat\u00f3rium sl\u00fa\u017ei na detekciu r\u00f4znych markerov za pou\u017eitia rozli\u010dn\u00fdch molekul\u00e1rno-biologick\u00fdch met\u00f3d ako napr. PCR\/qPCR\/dPCR, western blot, detekcia absorbancie, chemiluminiscencie a fluorescencie, \u010dipov\u00e1 kapil\u00e1rna elektrofor\u00e9za a pod. Molekul\u00e1rno-biologick\u00e9 laborat\u00f3ria s\u00fa vybaven\u00e9 kompletnou infra\u0161trukt\u00farou na izol\u00e1ciu DNA\/RNA\/prote\u00ednov, ich kvantifik\u00e1ciu (Nanodrop One) ako aj syst\u00e9mom pre sn\u00edmanie a vyhodnocovanie elektroforetick\u00fdch anal\u00fdz (ChemiDoc XRS+, Denzitometer GS-800).<\/p>\n

Laborat\u00f3rium analytickej cytometrie<\/strong><\/h4>\n

Laborat\u00f3rium je vybaven\u00e9 prietokov\u00fdm cytometrom BD FACSCalibur a sortrom BD FACSAria II SORP, ktor\u00fd umo\u017e\u0148uje multiparametrick\u00fa anal\u00fdzu fluorescencie r\u00f4znych \u010dast\u00edc (naj\u010dastej\u0161ie buniek) pripraven\u00fdch v suspenzii a ich n\u00e1sledn\u00e9 delenie na samostatn\u00e9 popul\u00e1cie na z\u00e1klade \u013eubovo\u013enej kombin\u00e1cie analyzovan\u00fdch parametrov. Takto je mo\u017en\u00e9 analyzova\u0165 a izolova\u0165 aj vz\u00e1cne sa vyskytuj\u00face elementy ako napr. kme\u0148ov\u00e9 bunky, n\u00e1dorov\u00e9 a nen\u00e1dorov\u00e9 bunky a pod. Medzi \u010fal\u0161ie vybavenie laborat\u00f3ria patria laborat\u00f3rne centrif\u00fagy s chladen\u00edm (Boeco, Labogene), platni\u010dkov\u00fd spektrofluoroluminometer (BMG FLUOstar Optima), NANODROP ONE (Thermoscientific), BioSpec-nano (Schimadzu), elektroforetick\u00e9 aparat\u00fary (Bio-rad PowerPaC Universal), molekul\u00e1rny zobrazovac\u00ed syst\u00e9m (Bio-rad ChemiDoc XRS+), PCR termocykl\u00e9r (Bio-rad C1000), RT-PCR termocykl\u00e9r (Bio-rad CFX96) a digit\u00e1lna PCR (dPCR).<\/p>\n

S\u00fa\u010das\u0165ou laborat\u00f3ria je fluorescen\u010dn\u00fd mikroskop DMI 6000 B (Leica Microsystems) s mikromanipul\u00e1torom a vysokov\u00fdkonnou metalhalidovou v\u00fdbojkou vhodnou na excit\u00e1ciu fluorof\u00f3rov v ultrafialovej, zelenej a \u010dervenej oblasti, ako aj sveteln\u00fdm zdrojom vhodn\u00fdm na sn\u00edmanie v jasnom poli (tzv. brightfield<\/em>). Mikroskop tie\u017e disponuje RGB filtrom vhodn\u00fdm na vyhodnocovanie kolokaliz\u00e1ci\u00ed. Mikroskop je vybaven\u00fd \u010diasto\u010dne motorizovan\u00fdm stol\u010dekom v rovine Z a \u0161tandardnou sadou objekt\u00edvov a vysokocitlivou \u010diernobielou kamerou. Konfok\u00e1lny mikroskop Leica TCS SP5 X vybaven\u00fd bielym laserom (tzv. WLL) a arg\u00f3nov\u00fdm laserom, ktor\u00fd je ur\u010den\u00fd na trojrozmern\u00fa mikroskopick\u00fa anal\u00fdzu a sledovanie dynamick\u00fdch zmien v \u010dase v bunk\u00e1ch a tkaniv\u00e1ch s mo\u017enos\u0165ou fluorescen\u010dnej spektr\u00e1lnej anal\u00fdzy. Mikroskop umo\u017e\u0148uje sn\u00edma\u0165 sn\u00edmky v jednej rovine, ako aj s\u00e9rie optick\u00fdch rezov (Z-stacky), tvorbu ortogon\u00e1lnych (X-Z a Y-Z) projekci\u00ed, projekciu maxim\u00e1lnej intenzity a kolokaliza\u010dn\u00e9 \u0161t\u00fadie na celul\u00e1rnej a subcelul\u00e1rnej \u00farovni.<\/p>\n

Laborat\u00f3rium v\u00fdvinovej neurobiol\u00f3gie<\/strong><\/h4>\n

Laborat\u00f3rium v\u00fdvinovej neurobiol\u00f3gie je zariaden\u00e9 infra\u0161trukt\u00farou, ktor\u00e1 umo\u017e\u0148uje vykon\u00e1va\u0165 izol\u00e1ciu tkan\u00edv, realiz\u00e1ciu imunohistochemick\u00fdch a imunofluorescen\u010dn\u00fdch zna\u010den\u00ed.
