Anot\u00e1cia: Regula\u010dn\u00e9 mechanizmy postnat\u00e1lnej neurogen\u00e9zy v neurog\u00e9nnej oblasti \u010duchov\u00e9ho syst\u00e9mu cicav\u010dieho mozgu – v subventrikul\u00e1rnej z\u00f3ne (SVZ), rostr\u00e1lnej migra\u010dnej dr\u00e1he (RMS) a bulbus olfactorius (BO) zost\u00e1vaj\u00fa st\u00e1le nedostato\u010dne vysvetlen\u00e9. Projekt je zameran\u00fd na sk\u00famanie regul\u00e1cie neurogen\u00e9zy prostredn\u00edctvom neurotransmiterov, oxidu dusnat\u00e9ho a seroton\u00ednu, v t\u00fdchto neurog\u00e9nnych oblastiach za fyziologick\u00fdch pomienok a na potka\u0148om modeli depresie. Na\u0161im cie\u013eom je odhali\u0165 neur\u00f3nov\u00e9 okruhy nitrergick\u00fdch neur\u00f3nov lokalizovan\u00fdch v RMS potkana a tak z\u00edska\u0165 cenn\u00e9 inform\u00e1cie o neuron\u00e1lnej regul\u00e1cii postnat\u00e1lnej neurogen\u00e9zy. S\u00favislosti medzi zn\u00ed\u017een\u00fdmi hladinami seroton\u00ednu v d\u00f4sledku depresie a zn\u00ed\u017eenou neurogen\u00e9zou boli demon\u0161trovan\u00e9 iba v hipokampe. Na\u0161e experimenty maj\u00fa za cie\u013e zisti\u0165 s\u00favislosti medzi depresiou, zmenenou regul\u00e1ciou seroton\u00ednu a neurog\u00e9nnymi procesmi v neurog\u00e9nnej oblasti \u010duchov\u00e9ho syst\u00e9mu. Anal\u00fdza integr\u00e1lnych komponentov mikroprostredia v SVZ u zvierat s indukovanou depresiou, umo\u017en\u00ed nahliadnu\u0165 do procesov regul\u00e1cie neurogen\u00e9zy za patologicky pozmenen\u00fdch podmienok. Nov\u00e9 poznatky v tejto oblasti by mohli odhali\u0165 n\u00e1dejn\u00e9 ciele pre nov\u00e9 terapie ochoren\u00ed s\u00favisiacich so zmenami v adultnej neurogen\u00e9ze.<\/p>\n

Anot\u00e1cia: Od rozsiahleho pilotn\u00e9ho skr\u00edningu pr\u00edrodn\u00fdch l\u00e1tok realizovan\u00e9ho National Cancer Institute v rokoch 1958 a\u017e 1980, ktor\u00fd vy\u00fastil do identifik\u00e1cie viacer\u00fdch \u00fa\u010dinn\u00fdch l\u00e1tok pr\u00edrodn\u00e9ho p\u00f4vodu, napr. paclitaxelu, uplynulo takmer 40 rokov. V\u00fdznam z\u00edskavania nov\u00fdch pr\u00edrodn\u00fdch l\u00e1tok pre terapiu, pr\u00edpadne diagnostiku n\u00e1dorov, napriek nov\u00fdm mo\u017enostiam imunoterapie a vyu\u017e\u00edvaniu syntetick\u00fdch prepar\u00e1tov v s\u00fa\u010dasnosti narast\u00e1. Jednou z pomerne m\u00e1lo presk\u00faman\u00fdch skup\u00edn pr\u00edrodn\u00fdch l\u00e1tok z h\u013eadiska ich biomedic\u00ednskeho potenci\u00e1lu s\u00fa antrachin\u00f3ny. V\u00fdnimku tvor\u00ed fotoakt\u00edvny naftodiantr\u00f3n hyperic\u00edn, ktor\u00e9ho farmakodynamick\u00fd potenci\u00e1l pre diagnostiku a terapiu n\u00e1dorov bol potvrden\u00fd mnoh\u00fdmi experiment\u00e1lnymi a klinick\u00fdmi \u0161t\u00fadiami. Tento projekt je zameran\u00fd na v\u00fdskum vybran\u00fdch antrachin\u00f3nov, ktor\u00e9 predstavuj\u00fa intermedi\u00e1ty v biosynt\u00e9ze hyperic\u00ednu v z\u00e1stupcoch rodu Hypericum <\/em>a vyskytuj\u00fa sa ako medziprodukty, resp. kone\u010dn\u00e9 produkty polyketidovej biosyntetickej dr\u00e1hy v endofytick\u00fdch izol\u00e1toch hostite\u013eov uveden\u00e9ho rastlinn\u00e9ho rodu. Projekt nadv\u00e4zuje na najnov\u0161ie v\u00fdsledky rie\u0161ite\u013esk\u00e9ho kolekt\u00edvu (Kim\u00e1kov\u00e1 et al. 2018), ktor\u00e9 vy\u00fastili do n\u00e1vrhu alternat\u00edvnej biosyntetickej dr\u00e1hy protin\u00e1dorov\u00e9ho lie\u010diva hyperic\u00ednu s k\u013e\u00fa\u010dov\u00fdm intermedi\u00e1tom skyr\u00ednom a \u010fal\u0161\u00edmi antrachin\u00f3nmi. Cie\u013eom projektu je i) na z\u00e1klade genomickej, transkriptomickej a metabolomickej anal\u00fdzy prispie\u0165 k poznaniu o regul\u00e1cii biosynt\u00e9zy \u0161tudovan\u00fdch antrachin\u00f3nov, o vz\u00e1jomnej komunik\u00e1cii rastlinn\u00fdch hostite\u013eov a ich endofytick\u00fdch mikroorganizmov vo vz\u0165ahu k sekund\u00e1rnemu metabolizmu s perspekt\u00edvou n\u00e1vrhu biotechnologickej produkcie; ii) determinova\u0165 ich potenci\u00e1lny (anti)genotoxick\u00fd, pr\u00edpadne DNA-protekt\u00edvny \u00fa\u010dinok a vplyv na dynamiku bunkov\u00e9ho cyklu a antioxida\u010dn\u00e9 vlastnosti a iii) uskuto\u010dni\u0165 pilotn\u00fa \u0161t\u00fadiu ich cytotoxick\u00e9ho vplyvu na n\u00e1dorov\u00e9 bunkov\u00e9 l\u00ednie a chorioalantoick\u00e9 membr\u00e1ny avi\u00e1rneho embrya a modula\u010dn\u00e9ho \u00fa\u010dinku na interakcie n\u00e1dorov\u00fdch a imunitn\u00fdch buniek s perspekt\u00edvnym vyu\u017eit\u00edm v protin\u00e1dororovej terapii.<\/p>\n

Anot\u00e1cia: Komplement\u00e1rna v\u00e4zba terapeutickej nukleovej kyseliny (TNA) na nefunk\u010dn\u00fa RNA je s\u013eubn\u00fd terapeutick\u00fd pr\u00edstup. Spolu s in\u00fdm typom TNA, ktor\u00fd sa via\u017ee na vybran\u00e9 prote\u00edny, ich m\u00f4\u017eeme skombinova\u0165 a vytvori\u0165 z nich nano\u010dasticu so synergick\u00fdm terapeutick\u00fdm \u00fa\u010dinkom. V s\u00fa\u010dasnosti je v\u0161ak cielen\u00fd transfer nano\u010dast\u00edc z nukleov\u00fdch kysel\u00edn (NANP) do buniek n\u00e1ro\u010dn\u00fd. Preto navrhujeme alternat\u00edvu, v ktorej sa \u0161pecifickos\u0165 NANP dosiahne podmienenou aktiv\u00e1ciou p\u00e1ru funk\u010dne vz\u00e1jomne z\u00e1visl\u00fdch nano\u010dast\u00edc. Tieto nanovl\u00e1kna s\u00fa jednotlivo nefunk\u010dn\u00e9, ale v pr\u00edtomnosti vybranej mRNA obidve vl\u00e1kna vz\u00e1jomne interaguj\u00fa pri\u010dom sa z nich uvo\u013e\u0148uj\u00fa funk\u010dn\u00e9 TNA. Na\u0161u koncepciu by sme chceli otestova\u0165 v in vitro podmienkach a na modeli bunkovej l\u00ednie, kde by mRNA vybran\u00e9ho onkog\u00e9nu aktivovala NANP, \u010d\u00edm by do\u0161lo k uvo\u013eneniu dvoch typov RNA- jedn\u00e9ho, ktor\u00fd stimuluje bunkov\u00fd imunitn\u00fd syst\u00e9m a RNA, ktor\u00e1 by inhibovala expresiu g\u00e9nov d\u00f4le\u017eit\u00fdch pre onkogen\u00e9zu. Synergick\u00fd \u00fa\u010dinok by naru\u0161il viabilitu buniek a z\u00e1rove\u0148 vyvolal imunitn\u00fa odpove\u010f.<\/p>\n

Anot\u00e1cia: Apopt\u00f3za predstavuje fyziologick\u00fd proces odumierania buniek bez naru\u0161enia stability okolit\u00e9ho tkaniva. Tento proces je d\u00f4le\u017eit\u00fd pre spr\u00e1vny v\u00fdvin nervov\u00e9ho syst\u00e9mu (NS) a udr\u017eanie homeost\u00e1zy v dospelosti. V d\u00f4sledku naru\u0161enia funkcie \u0161pecifick\u00fdch prote\u00ednov zapojen\u00fdch do apoptotick\u00fdch dr\u00e1h m\u00f4\u017ee nasta\u0165 porucha v\u00fdvinu r\u00f4znych \u0161trukt\u00far NS a naopak, nespr\u00e1vna regul\u00e1cia procesov apopt\u00f3zy m\u00f4\u017ee vy\u00fasti\u0165 do neurodegenerat\u00edvnych ochoren\u00ed. Problematick\u00e1 detekcia prote\u00ednov asociovan\u00fdch s apopt\u00f3zou, tzv. apoptotick\u00fdch markerov pomocou imunodetek\u010dn\u00fdch met\u00f3d prispieva k nedostato\u010dn\u00e9mu pokroku pri sk\u00faman\u00ed \u00faloh apopt\u00f3zy v NS po\u010das ontogen\u00e9zy, \u010di za patologicky pozmenen\u00fdch podmienok. Realiz\u00e1cia cie\u013eov projektu umo\u017en\u00ed charakterizova\u0165 \u010dasov\u00fa z\u00e1vislos\u0165 pr\u00edtomnosti k\u013e\u00fa\u010dov\u00fdch prote\u00ednov, ktor\u00e9 s\u00fa cie\u013eom imunodetek\u010dn\u00fdch met\u00f3d pri sk\u00faman\u00ed apopt\u00f3zy. Anot\u00e1cia: Hyperic\u00edn je zauj\u00edmavou pr\u00edrodnou l\u00e1tkou s v\u00fdrazn\u00fdmi fotosenzibiliza\u010dn\u00fdmi vlastnos\u0165ami, ktor\u00e9 maj\u00fa potenci\u00e1l vo fotodynamickej terapii a diagnostike. Hypoxick\u00e9 podmienky be\u017ene pr\u00edtomn\u00e9 v n\u00e1doroch, \u010di aktivita transport\u00e9ra BCRP v\u0161ak m\u00f4\u017eu zna\u010dne ovplyvni\u0165 akumul\u00e1ciu hyperic\u00ednu v n\u00e1dorov\u00fdch bunk\u00e1ch, a t\u00fdm zni\u017eova\u0165 efektivitu terapie. Mechanizmus t\u00fdchto interakci\u00ed, \u010di \u0161pecifick\u00e9 izoformy BCRP zodpovedn\u00e9 za transport hyperic\u00ednu v\u0161ak neboli doteraz pop\u00edsan\u00e9 a bud\u00fa preto predmetom aktu\u00e1lneho projektu. Na\u0161e predbe\u017en\u00e9 v\u00fdsledky tie\u017e nazna\u010dili komplexnos\u0165 vplyvu nefotoaktivovan\u00e9ho hyperic\u00ednu na n\u00e1dorov\u00e9 bunky. Z h\u013eadiska protin\u00e1dorovej terapie je zvl\u00e1\u0161\u0165 zauj\u00edmav\u00fd jeho potenci\u00e1lny vplyv na angiogen\u00e9zu, prote\u00edny zapojen\u00e9 do priestorovej organiz\u00e1cie a adhez\u00edvnosti, ako aj schopnos\u0165 modulova\u0165 ve\u013ekos\u0165 a vlastnosti rezistentnej, tzv. bo\u010dnej popul\u00e1cie buniek. V predkladanom projekte sa preto zameriame aj na uveden\u00e9 \u00fa\u010dinky hyperic\u00ednu, ktor\u00e9 budeme sledova\u0165 in vitro, zahypoxie, ale aj na modeli chorioalantoickej membr\u00e1ny.<\/p>\n

Anot\u00e1cia: Side population<\/em> (SP) predstavuje met\u00f3du prietokovej cytometrie, ktorou dok\u00e1\u017eeme identifikova\u0165 tzv. SP popul\u00e1ciu, a to na z\u00e1klade zv\u00fd\u0161en\u00e9ho efluxu prostredn\u00edctvom ABC transportn\u00fdch prote\u00ednov. SP bunky tak reprezentuj\u00fa rezistentn\u00fa frakciu v n\u00e1doroch, a t\u00fdm aj potenci\u00e1lny cie\u013e protin\u00e1dorov\u00fdch strat\u00e9gi\u00ed. Nako\u013eko v\u0161ak inhib\u00edtory ABC transportn\u00fdch prote\u00ednov nepreuk\u00e1zali signifikantn\u00fd pr\u00ednos v klinickej praxi, je d\u00f4le\u017eit\u00e9 identifikova\u0165 aj in\u00e9, alternat\u00edvne molekuly, ktor\u00e9 s\u00fa selekt\u00edvne pr\u00edtomn\u00e9 v SP bunk\u00e1ch a mohli by by\u0165 objektom alternat\u00edvnych protin\u00e1dorov\u00fdch pr\u00edstupov. Cie\u013eom projektu je komplexn\u00e1 proteomick\u00e1 anal\u00fdza a porovnanie prote\u00f3mu SP a nonSP popul\u00e1ci\u00ed izolovan\u00fdch z bunkovej l\u00ednie p\u013e\u00facneho karcin\u00f3mu A549. Pr\u00ed\u010dina vzniku n\u00e1dorovej hypoxie, mechanizmus odpovede buniek na hypoxiu a celkov\u00fd dopad hypoxie na priebeh onkologickej lie\u010dby s\u00fa z\u00e1kladn\u00fdmi ot\u00e1zkami, ktor\u00e9 si mus\u00edme zodpoveda\u0165, ak chceme detailnej\u0161ie porozumie\u0165 mechanizmu progresie n\u00e1dorov\u00e9ho ochorenia. V s\u00fa\u010dasnosti je v\u00fdskum zacielen\u00fd na h\u013eadanie inhib\u00edtorov sign\u00e1lnych dr\u00e1h zapojen\u00fdch do bunkovej odpovede na chemoterapiu.
